The determination is made from a venous blood sample from the arm.

It will depend on the type of sample requested by the doctor, be it saliva, urine, or blood.
Why do the analysis?
To facilitate the diagnosis of Cushing's syndrome or primary (Addison's disease) or secondary adrenal insufficiency; to detect disorders affecting the pituitary gland or adrenal glands.
When to do the analysis?
When there is suspicion of excess or deficit in cortisol production.
